The disease causes lesion (canker), wilting and dryness of plants, leaf and fruit spots and whole plant decline. In this study, about 160 samples of infected plants were collected and sixty-four bacterial isolates were obtained from the infected tomato plants in Golestan and West Azarbaijan during 2004-2005. All isolates identified as Clavibacter michiganensis subsp. michiganensis based on morphological biochemical characters and pathogenecity test. Isolate Cmm (NCPPB382, Germany) was used as standard. Isolates showed considerable differences in using different sources of carbon and in their electrophoretic (SDS-PAGE) profiles of soluble cell proteins. Cluster analysis dendrograms obtained based on information derived from biochemical characters and protein electrophoresis, using software NTSYS-pc version 2/o2e, UPGMA method and Jaccard′s coefficient. Three groups (clusters) with %77 similarity based on biochemical data, 5 groups with %86 similarity based on SDS-PAGE results were realized. The results indicated high correlation and complementarirty. Resultes revealed I) heterogenesis between Iranian isolates and standard Cmm (NCPPB382), II) homogenesis between Golestan and West Azarbaijan isolates, and III) presence of different populations (strains) among the isolates selected from different areas. This is the first report of tomato bacterial disease in Golestan provice of Iran.}, keywords = {Clavibacter michiganensis subsp. michiganensis,Bacterial canker of tomato,West Azarbaijan, Golestan province}, title_fa = {بررسی تنوع فنوتیپی و پروتئینی عامل بیماری شانکر باکتریایی گوجه‌فرنگی در استان‌‌های گلستان و آذربایجان غربی}, abstract_fa = {بیماری شانکر باکتریایی گوجه‌فرنگی در ایران، اولین بار در سال 1367 از منطقه ارومیه در استان آذربایجان غربی گزارش گردید. این بیماری موجب زخم (شانکر)، پژمردگی و خشکیدگی یک طرفه در بوته و ایجاد لکه روی برگ و میوه شده و در نهایت به زوال کامل گیاه منجر می‌گردد. در طی این تحقیق، 64 جدایه باکتریایی از بوته‌‌های آلوده گوجه فرنگی مزارع استان آذربایجان غربی و گلستان جداسازی و بررسی گردید. بر اساس مشخصات مرفولوژیکی، بیوشیمیایی و خصوصیات بیماری‌زایی، تمامی‌ جدایه‌‌ها تحت عنوان Clavibacter michiganensis subsp. michiganensis شناسایی شدند. در تمامی ‌مراحل شناسایی از نمونه استاندارد Cmm (NCPPB382، از آلمان) به عنوان شاهد در آزمون‌‌ها استفاده گردید. نتایج، بیانگر وجود تفاوت‌‌های مشخص در توانایی استفاده از منابع مختلف و نقوش الکتروفورتیکی پروتئین محلول سلولی با استفاده از روش SDS-PAGE بود. دندروگرام داده‌‌های حاصل از خصوصیات بیوشیمیایی و الکتروفورز پروتئین با استفاده از نرم افزارNTSY`S-pc Version 2/02e، روش UPGMA و ضریب جاکارد رسم گردید. تعداد سه گروه با درصد شباهت معادل 77% در دندروگرام حاصل از داده‌‌های بیوشیمیایی و پنج گروه با درصد شباهت معادل 86% در دندروگرام حاصل از داده‌‌های پروتئینی مشخص گردید. نتایج فوق هتروژنی در بین جدایه‌‌های ‌ایرانی با نمونه استاندارد، هموژنی بین جدایه‌‌های گلستان و آذربایجان غربی و وجود جمعیت‌‌های متفاوت در بین جدایه‌‌های مناطق مختلف را نشان دادند. این اولین گزارش از وجود بیماری شانکر باکتریایی گوجه فرنگی در استان گلستان است.}, keywords_fa = {Clavibacter michiganensis subsp. michiganensis,شانکر باکتریایی گوجه‌فرنگی,ایران,آذربایجان غربی,گلستان}, url = {https://jaenph.areeo.ac.ir/article_109476.html}, eprint = {https://jaenph.areeo.ac.ir/article_109476_ca193ae270c44d35c9e797821c3260c3.pdf} }
