Investigation of Extract of Several Seaweeds on Potato Virus Y in Tobacco

Abstract

Tobacco (Nicotiana tabacum L.) is an annual plant from the family of Solanaceae whose products are widely used in the pharmaceutical industry and commerce. One of the most important tobacco viruses which have a global distribution is the Potato Virus Y (PVY). However, there has not been a method presented for controlling the disease except introducing the resistance cultivar. In order to investigate the effect of different concentrations of Aqueous and Methanolic extracts of three algae species (Digenea simplex (Wulfen) C.Agard, Gracilaria corticata (J.Agardh) J.Agardh and Padina australis) on the intensity and ELISA absorbance (OD) of Potato Virus Y (Accession: MF688631.1) and tobacco chlorophyll content, an experiment in a completely randomized design with four replications were performed in 2019-2020 in Greenhouse conditions. One month after virus infection the leaflets at the top of each plant were picked in order to determine from the ELISA. The severity of virus and the amount of chlorophyll were measured. The results showed that the algae type and extract type had a significant effect on the severity of symptoms, the final concentration of the virus and the amount of chlorophyll in the tobacco plant. In this study, Padina australis Hauck showed the greatest effect on reducing the virus concentration (55.06%) and the severity of disease symptoms. The highest levels of chlorophyll were also observed in this treatment. In the study of extraction methods, Methanolic extracts showed better performance than aqueous extracts.
