Citrus yellow vein clearing virus (CYVCV) is an emerging citrus pathogen that has spread to several countries, including north of Iran, over the past two decades. During 2023, the typical symptoms of the citrus yellow vein clearing disease including stunted growth, and vein clearing preceded by a water soaked appearance on the lateral veins of the abaxial leaf surface was observed in sour orange trees in an urban area in Shiraz (Fars Province). To detect the virus, reverse transcription polymerase chain reaction (RT-PCR) was conducted using a specific primer pair targeting the coat protein (CP) gene. An approximately 610 bp fragment of the coat protein gene was amplified from symptomatic sour orange samples but not from the samples of healthy trees. Sequencing of one PCR product and a BLASTn search of the resulting sequence against the GenBank (NCBI) database revealed the highest identity to sequences of Citrus yellow vein clearing virus isolates. This is the first report of the occurrence of citrus yellow vein clearing disease in Fars Province. The virus may be present in other parts of southern Iran, which calls for expanded diagnostic screening and monitoring.
