تنوع ژنتیکی و هیبریداسیون احتمالی بین فرم‌های اختصاصی زنگ زرد جو و گندم در ایران با استفاده از نشانگرهای SSR و IGS1

چکیده

بیماری زنگ زرد غلات با عامل Puccinia striiformis یکی از مهم‌ترین بیماری‌های اقتصادی در ایران و جهان است. این بیمارگر دارای فرم‌های اختصاصی مختلف از جمله فرم اختصاصی گندم (Pst) و جو (Psh) می‌باشد. تولیدمثل جنسی روی زرشک، هیبریداسیون و مهاجرت‌های بین‌قاره‌ای در تنوع ژنتیکی بالای آن، به‌ویژه در جمعیت‌های آسیایی، نقش کلیدی دارند. در این پژوهش، تنوع ژنتیکی ۱۸ جدایه (نماینده ۳۶ جدایه) شامل ۱۲ جدایه Psh از جو و ۶ جدایه Pst از گندم و جو وحشی، از مناطق مختلف ایران، با استفاده از ناحیه IGS1 و پنج نشانگر ریزماهواره‌ای (RJ18, RJ21, RJ2N, RJ10N, RJ11N) بررسی شد. نشانگر IGS1 با هفت الگوی باندی، جدایه‌های گندم را از جو به خوبی متمایز کرد. در نشانگرهایSSR، ۱۴ آلل با میانگین ۸/۲ آلل در هر جایگاه شناسایی شد و جایگاه RJ2N با ۴ آلل و بالاترین شاخص PIC (۰٫۴)بیشترین تنوع را نشان داد. تنوع ژنتیکی در جدایه‌های Psh بیشتر از Pst بود. الگوهای باندی دوگانه در برخی جدایه‌های Psh نشان‌دهنده آلل‌های مشترک با Pst و احتمال هیبریداسیون بود. دندروگرام UPGMA جدایه‌ها را در سه گروه اصلی قرار داد. شباهت ژنتیکی بین جدایه‌های مناطق دور از هم، بیانگر جریان ژنی فعال است. تنوع بالای مشاهده‌شده در جمعیت‌های ایرانی، به‌ویژه در اردبیل، نشان‌دهنده پتانسیل تکامل سریع بیمارگر و ضرورت پایش مستمر با ابزارهای مولکولی دقیق‌تر است. شناسایی جدایه‌های هیبرید زنگ خطری جدی برای کشاورزی است، زیرا هیبریداسیون می‌تواند با انتقال ژن‌های بیماری‌زایی و تهاجمی بین فرم‌های اختصاصی، به ظهور نژادهای جدید با خسارت بیشتر و شکست مقاومت ارقام منجر شود.

Stripe rust of cereals is one of the most economically important diseases in Iran and worldwide. This pathogen has various forma specialis, including the wheat form (Pst) and the barley form (Psh). Sexual reproduction on barberry, hybridization, and intercontinental migrations play key roles in its high genetic diversity, especially in Asian populations. In this study, genetic diversity of 18 isolates (representing 36 isolates), comprising 12 Psh isolates from barley and 6 Pst isolates from wheat and wild barley collected from different regions of Iran, was investigated using the IGS1 region and five microsatellite markers . The IGS1 marker clearly distinguished wheat from barley isolates by generating seven distinct banding patterns. SSR analysis detected 14 alleles with an average of 2.8 alleles per locus; the RJ2N locus exhibited the highest diversity with 4 alleles and the highest PIC value (0.4). Genetic diversity was higher in Psh than in Pst isolates. Double banding patterns in some Psh isolates indicated shared alleles with Pst and possible hybridization. The UPGMA dendrogram grouped isolates into three main clusters. Genetic similarity among isolates from distant regions suggests active gene flow. The high diversity in Iranian populations, especially in Ardabil, highlights the potential for rapid pathogen evolution and underscores the need for continuous monitoring using more precise molecular tools. Identification of hybrid isolates is a serious warning for agriculture, as hybridization can lead to emergence of new, more damaging races by transferring pathogenicity and aggressiveness genes between forma specialis, ultimately resulting in breakdown of host resistance
