چکیده

شارکا یا بیماری آبله‌ی آلو بوسیله Plumpoxvirus (PPV) عضو جنس Potyvirus ایجاد می‌شود (1). این بیماری از لحاظ اهمیت اقتصادی یکی از مخرب‌ترین بیماری‌های درختان میوه هسته‌دار می‌باشد. کاهش کیفیت میوه‌ها و ریزش پیش از موقع آنها سبب شده است که این بیماری یکی از فاکتورهای محدود کننده در تولید هسته‌داران خصوصاً درختان زردآلو، هلو و آلو به حساب آید. بیماری آبله‌ی آلو در قسمت‌های وسیعی از قاره اروپا، حوزه مدیترانه و برخی از کشورهای خاورمیانه گسترش یافته است.
در اواخر تابستان و پائیز سال 1389 به دنبال مشاهده ریزش شدید میوه آلو مشکوک به آلودگی به PPV در برخی از باغ‌های آلو در استان مازندران، 75 نمونه برگ و شاخه جمع آوری و بررسی گردید. نمونه برداری بر اساس علائم شاخص PPV انجام شد و آلودگی ویروسی توسط آزمایش‌های سرولوژیکی و مولکولی مورد تائید قرار گرفت. در تشخیص سرولوژیکی نمونه‌ها، از آزمون ساندویچ دو طرفه الایزا (DAS-ELISA) با آنتی‌سرم‌های چند همسانه‌ای اختصاصی PPV (تهیه شده از شرکت Bioreba سوئیس) استفاده شد. تشخیص مولکولی بوسیله آزمایش IC-RT-PCR با بدام اندازی پیکره ویروسی با آنتی سرم فوق و با استفاده از جفت آغازگر P1/P2 انجام شد. نتایج نشان داد که 48 نمونه از 75 نمونه مورد آزمایش به ویروس آبله‌ی آلو آلوده بودند. به منظور تائید نتیجه بدست آمده، آر ان ای کل نمونه‌های مثبت بوسیله کیت استخراج RNA شرکت کیاژن مطابق با دستورالعمل از بافت جوانه‌های در حال خواب و پوست شاخه استخراج گردید (2). واکنش زنجیره‌ای پلیمراز با ترانویسی معکوس یک مرحله‌ای با استفاده از Qiagene One-Step RT-PCR Kit، وجود قطعه 243 جفت بازی مورد انتظار را که در قسمت انتهای کربوکسیلایی ژن پروتئین پوششی ویروس قرار گرفته است(3)، در تمامی 48 نمونه مورد آزمایش به تائید رساند. نتایج آزمون واکنش زنجیره‌ای پلیمراز با ترانویسی معکوس با آزمون‌های DAS-ELISA و IC-RT-PCR کاملا مطابقت داشت.
از آنجایی که شدت علائم تا حد زیادی به گونه گیاهی و استرین ویروس بستگی دارد، بررسی مولکولی نوع استرین نیز با استفاده از جفت آغازگر‌های P1/PD و P1/PM که به ترتیب دو استرین مهم PPV را تشخیص می‌دهد، صورت گرفت. در آنالیز PCR-RT انجام شده مشخص گردید که هر 48 نمونه به استرین نوعM  آلوده بوده و استرین نوع D و یا آلودگی توام در هیچ یک از ایزوله‌ها شناسایی نگردید. متعاقباً، نوع استرین، بر اساس آنالیز چند شکلی طول قطعات برشی (RFLP) با استفاده از اندونوکلئازهای برشی RsaI و AluI روی قطعه 243 جفت بازی تکثیر شده، مورد تائید قرار گرفت (4). در این مطالعه نقوش حاصل از هضم آنزیمی محصول PCR– RT در تمامی نمونه‌های مورد آزمایش تنها شامل سایت برشی AluI بودند.

Association of Plum Pox Virus Strain M with Plum Fruit Dropping in Mazandaran Province (short report)

چکیده [English]

Sharka (plum pox disease) is caused by Plum pox virus (PPV) a member of the genus Potyvirus (1). It is considered one of the most devastating diseases of stone fruits in terms of economic importance. The disease causes reduced quality and premature dropping of fruit and is one of the limiting factor of apricot, peach, plum and certain other stone fruit production in Iran. The disease has progressively spread to a large part of the European continent, Mediterranean basin and some of Middle East countries.
Severe fruit dropping was recently observed in some plum orchards of Mazandaran province. In order to investigate possible involvement of PPV, 75 leaf and shoot samples collected during the late summer and autumn of 2010. Sampling was based on typical PPV symptoms. Virus infection was confirmed by serological and molecular testing. Serological diagnosis was made by DAS-ELISA using a commercial PPV polyclonal antiserum (Bioreba, Switzerland). Molecular detection was made by trapping virus particle with the above polyclonal antiserum and IC-RT-PCR was performed by using the general pair of primers P1/P2. Total RNA were extracted from dormant buds and barks (2) using RNeasy Plant Mini Kit (Qiagen, Germany) according to the manufacturer’s instructions.
The results showed that 48 out of 75 plum samples were found to be infected with PPV. One-step RT-PCR (Qiagene One-Step RT-PCR Kit) was performed using the general primer pair (P1/P2). The P1/P2 primers revealed and confirmed the presence of the virus by amplifying the expected 243 bp fragment located at the C-terminus of PPV CP gene (3). The results of RT-PCR analysis were in complete agreement with the DAS-ELISA and IC-RT-PCR results.
Since, the severity of symptoms depends largely on plant species and virus strain, the type of strain determined by RT-PCR targeting (Cter) CP, using P1/PD and P1/PM pair of primers that distinguish two major and PPV-D strains PPV-M, respectively; The RT-PCR analyses confirmed that all 48 samples were infected with PPV-M type and no samples were positive for PPV-D. Subsequently, molecular strain typing was confirmed by Restriction Fragment Length Polymerase (RFLP) of 243 bp amplicon using RsaI and AluI restriction endonucleases digestion (4). In this study, the pattern after enzyme digestion showed all of PCR products contained the AluI site.
