چکیده

باکتری Pseudomonas syringae pv. syringae (Pss)عامل یکی از بیماری‌های شایع در باغ‌های درختان میوه هسته‌دار می‌باشد. به منظور شناسایی جدایه‌های عامل بیماری، طی سال‌های 1391-1390، 23 جدایه به‌دست آمده از باغ‌های درختان میوه هسته‌دار واقع در مناطق مختلف استان‌های خراسان رضوی و شمالی از نظر فنوتیپی (فیزیولوژیکی و بیوشیمیایی) و بیماری‌زایی مورد بررسی قرار گرفتند. جدایه‌ها از نظر ویژگی‌های فنوتیپی اختلافات جزئی نشان دادند. همه جدایه‌ها روی هلو و بادام دارای قدرت بیماری‌زایی بالایی بودند و از این نظر مشابه به نظر رسیدند. صحت تشخیص جدایه‌ها در سطح گونه توسط واکنش زنجیره‌ای پلیمراز و با استفاده از آغازگر‌های D21/D22 اختصاصی گونه مورد تایید قرار گرفت. تنوع ژنتیکی جدایه‌ها به دو روشrepetitive element palindromic PCR (rep-PCR) و با استفاده از آغازگر‌های BOXA1R، ERIC1R و ERIC2 و روش Insertion Sequence PCR (IS-PCR) و با استفاده از آغازگر IS50 مورد بررسی قرار گرفت. آنالیز خوشه‌ای نتایج به دست آمده نشان داد که در BOX-PCR با آغازگر BOXA1Rدر سطح تشابه 75 درصد، جدایه‌ها به 7 گروه؛ در ERIC-PCR با آغازگر ERIC1R در سطح تشابه 65 درصد، جدایه‌ها به 7 گروه و با آغازگر ERIC2 در سطح تشابه 56 درصد، جدایه‌ها به 8 گروه؛ در IS-PCR با آغازگر‌IS50 در سطح تشابه 52 درصد، جدایه‌ها به 11 گروه و در نهایت با ترکیب هر چهار آغازگر‌ در سطح تشابه 54 درصد جدایه‌ها به 7 گروه قابل تفکیک هستند. نتایج بیانگر تنوع ژنتیکی بالا درون جدایه‌های عامل بیماری شانکر باکتریایی درختان میوه هسته‌دار در استان‌های مورد مطالعه است.

چکیده [English]

Pseudomonas syringae pv. syringae (Pss) is the causative agent one of the bacterial common diseases in stone fruit trees fields. In this research, samples were collected from various areas in Razavi and Northern Khorasan provinces during 2011-2012. A total of 23 bacterial isolates were compared based on their phenotypic (physiological and biochemical) characteristics and pathogenicity tests. Pss strains showed slight differences in phenotypic properties. All 23 strains were highly pathogenic on almond and peach seedlings. All strains appeared to be similar in pathogenicity traits. Identification of the strains was further confirmed by PCR using D21/D22 primers specific to P. syringae. Genetic diversity of selected strains was investigated by repetitive element palindromic PCR (rep-PCR) fingerprinting using BOXA1R, ERIC1R, ERIC2 primers and Insertion Sequence PCR (IS-PCR) fingerprinting using IS50 primers. The cluster analyses of the fingerprint data showed that in BOX-PCR with BOXA1R primer, 7 groups were developed at a similarity level of 75%. The same number of groups were obtained in ERIC-PCR using ERIC1R primer at a similarity level of 65%. In case of ERIC2 primer, at a similarity level of 56%, strains were divided into 8 groups, whereas, in IS-PCR with IS50 primer at a similarity level of 52%, strains were divided into 11 groups and finally by combining all four primers at a similarity level of 54%, strains can be distinguished into 7 groups. The results demonstrated the existence of a considerable genetic diversity among Pss strains causing bacterial canker in stone fruit trees in the studied provinces.
