چکیده

سرخشکیدگی درختان پسته یکی از مهمترین و شایع­ترین بیماری‌های درختان پسته می­باشد که به لحاظ اقتصادی خسارت زیادی به صنعت پسته کشور وارد آورده است. با توجه به نقش باکتری­ها در بروز و توسعه این بیماری، برهمکنش بین جدایه‌های Bacillus spp. و قارچ Paecilomyces formosus در ایجاد و توسعه بیماری بررسی شد. به این منظور ابتدا توان بیماری‌زایی جدایه ها روی سرشاخه‌ در شرایط آزمایشگاه ارزیابی، و جدایه‌های منتخب در شرایط باغ زیر بافت پوست سرشاخه‌های پسته رقم اوحدی مایه­زنی شد. براساس نتایج به‌دست آمده، جدایه‌های Bacillus pumilus و P. formosus به تنهایی قادر به ایجاد بیماری روی سرشاخه‌های درختان پسته در شرایط باغ بودند که میزان پیشروی طولی علائم در بافت آوندی در مایه­زنی تنها با جدایه­های باکتریایی از 4.5 تا 14 و در ترکیب با جدایه قارچی از 10 تا 32 سانتی‌متر متغیر بود. در سرشاخه­های مایه­زنی­شده، جداسازی جدایه‌های باکتریایی از مرز علائم و 10 سانتی­متر پایین­تر از آن در بافت سالم سرشاخه نیز صورت گرفت. براساس ویژگی­های مرفولوژیکی، بیوشیمیایی و مولکولی، جداسازی جدایه‌های باکتریایی B. pumilus برای اولین‌بار به‌عنوان یکی از عوامل سرخشکیدگی درختان پسته گزارش می­شوند.

Pistachio die-back is one of the most important and prevalent diseases of pistachio orchards, causing severe reduction of pistachio yield. Due to the important role of bacteria in the occurrence and development of pistachio die-back, the interaction of Bacillus spp. and Paecilomyces formosus was investigated in this research. The isolates of both groups were injected beneath of the inner bark of branches (Ohadi cultivar) using sterile syringes alone or in mixture. According to the results, Bacillus pumilus and P. formosus isolates were able to cause disease on the branches, with longitudinal developments in vascular tissue. Bacterial isolates alone caused lesions ranged 4.5-14 cm in inoculation sites. Mix inoculations, significantly, increased disease development in xylem varied 10-32 cm. Based on morphological, biochemical and molecular properties, isolation of B. pumilus strains is reported for the first time as one of the causal agents of pistachio die-back in Iran.
